The present investigation aimed at development and validation of an analytical method for the estimation of Gemcitabine Hydrochloride based on Ultra-violet spectrophotometry. The reported methods of analysis include the chromatographic methods such as HPLC (1) RP-HPLC (2-4), Ion pair HPLC (5), and LC-MSMS (6). Chromatographic methods are tedious, time consuming and are not suitable for routine analysis. However, Ultraviolet spectophotometric methods which are simple and fast are ideal for routine analysis. Gemcitabine Hydrochloride exhibits a sharp peak at 268 nm when scanned in the Ultraviolet region. Thus, it was selected as the analytical wave-length. The developed method was found to be simple, sensitive, accurate, precise and specific as evidenced by non-interference from the excipients present in the marketed injection. Linearity was observed in the range of 5µg/ml to 35 µg/ml. The limit of detection (LOD) and limit of quantitation (LOQ) were found to be 0.384 µ/ml & 1.163 µ/ml respectively....
A simple, rapid, accurate, precise and economic RP-HPLC method was developed for the estimation of aprepitant in bulk and pharmaceutical dosage forms using C18 column (250X 4.6 mm i.d; 5µm) in isocratic mode, with mobile phase comprising of methanol and water in the ratio of 90:10 (v/v). The flow rate was maintained 1 ml/min and the detection was carried out at 220 nm with variable wavelength detector. The retention time (tr) of aprepitant and the rimonabant hydrochloride (IS) was found to be 4.383 and 5.783 min, respectively. The method has permitted linear response in the concentration range of 1- 100 µg/ml and the limit of detection and limit of quantification was found to be 0.130 and 0.395, respectively. The percentage recovery was found to be 99.56 to 101.5 and the % RSD of system precision and method precision was found to be 1.20 and 0.561, respectively. Rimonabant hydrochloride was used as an internal standard in the present study. An experiment was conducted to develop a drying protocol for Withania somniferum during the year 2009-10 under National Medicinal Plant Board project, B.R. Nahata college of pharmacy- SIRO (scientific institute of research organization), Mandsaur (M.P.). The roots of withania were harvested and cut into different sizes (2-4 inches) and subjected into sun drying, shade drying, and hot air oven drying in temperature ranging from 40-600C. Temperature for sun drying ranging from 32-420C during day time and in night hours, the samples were kept in plastic covers to prevent re absorption of moisture. In shade drying pieces of roots were kept in shade (proper air flowing place) at normal climatic temperature. And in hot air oven drying method/ mechanical drying method the pieces of roots were kept at 500C, 550C and 600C respectively. The samples were weighed and moisture content of the samples were taken at regular intervals till the standard moisture content of the dried roots was obtained that is mentioned in literatures (6% or below 6%). The result revealed that hot air oven drying of roots took the lowest time (12hrs.). while sun drying method took 24hrs for drying. The alkaloid content viz., withanolide A was significantly influenced by different methods of drying. The highest alkaloid content (0.021%) was observed in hot air oven drying as compared to sun drying with 0.010% and shade drying with 0.009% Withanolide A....
In the present study bilayered tablet coating of two drugs were prepared which are complimentary of each other. One is glyburide which was used for quick release and the other is Metformin Hydrochloride which was used for sustained release using super disintegrant property of sodium starch glycolate for latter and matrix forming agents like Hydroxyl Propyl Methyl Cellulose (HPMC K 4) and Sodium Carboxy ethyl Cellulose (SCMC) and binder like PVPK-30 for former. All the tablets were evaluated for their physicochemical properties and were found within satisfactory region. Regression coefficient method was used for evaluation of the release kinetics of Metformin HCl. Various formulations were made so as to get a good release profile out of which only one (F4) showed zero order release in which dominant mechanism was diffusion. Because of the significant effect of the polymers which were used it was possible to attain quick release of glyburide and sustain release of Metformin hydrochloride over a period of approximately 8 hours....
Simple, sensitive and specific Chromatographic method was developed and validated for simultaneous estimation of Diacerein (DIA) and Aceclofenac (ACE) in tablet dosage form. The new analytical method was developed based on the simultaneous estimation of drugs in a binary mixture without previous separation. In graphical vierordt’s method was performed by absorbance at 254.0nm and 298.0nm. Both the drug DIA and ACE and its mixture follow beer-lamber’s law in the range of 2 -10 μg/ml at the entire selected wavelengths. The percent estimation of mix drug in laboratory mixture was done to be 100.57 + 0.64 and 99.55 + 1.74 for DIA and ACE respectively. The percent drug estimation in marketed formulation was found to be 100.82+ 0.91 and 99.61 +1.46 the average percent recovery was found to be 99.28 +1.30 and 98.54+0.56. The result of the method lies within the prescribe limit of 98-102% shows that method is free from interference from excipients....
Two simple, accurate, economical and reproducible UV Spectrophotometric methods for simultaneous analysis of Terfenadine and Dextromethorphan hydrogen bromide from combined suspension dosage form have been developed. First method was developed making use of fourth order derivative spectroscopy using 274.8 nm and 298.0 nm as zero crossing points for estimation of Terfenadine and Dextromethorphan hydrogen bromide respectively. Second developed method is based on two wavelength calculation; wavelengths selected for estimation of Terfenadine were 220.6 nm and 230.0 nm. Estimation of Dextromethorphan hydrogen bromide was carried out directly at 280.0 nm as Terfenadine has zero absorbance at this wavelength. Solvent used for development of Spectrophotometric methods was methanol. The results of analysis were validated statistically and by recovery studies....
